目的:观察芪桂痛风片对痛风性关节炎大鼠、肉芽肿大鼠、耳肿胀小鼠的抗炎作用。方法:灌胃给予芪桂痛风片5 d后,采用大鼠右后踝关节腔内注射尿酸钠混悬液,制备急性痛风性关节炎模型,测量造模后1h、3h、5h的大鼠足容积,并对踝关节组织进行病理组织学检查;同时采用二甲苯致小鼠耳肿胀模型、纽扣植入引起的大鼠肉芽增生模型,观察芪桂痛风片的抗炎作用。结果:芪桂痛风片3个剂量组均能明显降低急性痛风性关节炎大鼠给药后3h、5h的足肿胀度和肿胀率,病理组织学检查显示,给药后关节组织水肿和炎性细胞浸润、变性坏死有不同程度的减轻,8.10g/kg组病变评分明显低于模型组。芪桂痛风片8.10g/kg、4.05g/kg组肉芽组织湿重、干重均比模型组明显减轻。芪桂痛风片11.70g/kg、5.85g/kg组小鼠耳廓肿胀度及肿胀率与模型组比较明显降低。结论:芪桂痛风片可降低急性关节炎模型大鼠的关节肿胀,减轻关节病理变化的程度,抑制肉芽组织生长、小鼠耳廓肿胀,具有较强的抗炎作用。芪桂痛风片对大鼠的无效剂量是2.03g/kg,有效剂量是4.05g/kg;对小鼠无效剂量是2.93g/kg,有效剂量是5.85g/kg。
