目的进一步了解长春市地区牧场周围污染的土壤、水中非结核分枝杆菌(NTM)分布情况。方法对6株分离自吉林地区牧场周围污染的水及土壤中的疑似NTM菌株进行基因组提取,并通过PCR方法成功扩增待检NTM菌株16S rDNA基因序列,通过GenBank对6株待检菌16S rDNA基因序列进行blast分析比较鉴定。结果长春市周边牧场环境中的水和土壤各采集300份样品,经过抗酸染色初步鉴,6株菌株为阳性,经过Blast分析发现所测6株待测菌中,新金分枝杆菌3株,耻垢分枝杆菌1株,胞内分枝杆菌1株,浅黄分枝杆菌1株。结论长春市周边牧场环境中非结核分枝杆菌的分离率为2.0%。应对牧场周边进一步加强管理和监督,预防非结核分枝杆菌。
Objective To understand the distribution of non-tuberculous mycobacterium (NTM) in contaminated soil and water in the pasture around Changchun City. Methods Genomic DNA was extracted from 6 suspected NTM isolates from contaminated water and soil in the pasture around Jilin Province. The 16S rDNA gene sequence of NTM strain was successfully amplified by PCR. Six strains of 16S rDNA Blast analysis and identification of gene sequences. Results 300 samples were collected from the water and soil in pasture environment around Changchun City. Six samples were positive after acid-fast staining. Blast analysis showed that there were 3 strains of Mycobacterium phlei 1 strain of Mycobacterium smegmatis, 1 strain of Mycobacterium intracellulare and 1 strain of Mycobacterium bovis. Conclusion The isolation rate of Mycobacterium tuberculosis in pasture environment around Changchun City was 2.0%. We should further strengthen the management and supervision around the ranch to prevent non-tuberculous mycobacteria.
