目的:探讨丹蛭降糖胶囊对大鼠糖尿病肾病的防治作用,为其临床应用提供实验依据。方法:雄性SD大鼠80只,随机选取12只作为正常对照组,其余大鼠以高脂饲料+链脲佐菌素(STZ)(34mg/kg腹腔注射)造模,选取空腹血糖≥7.0mmol/L同时剔除血糖过高大鼠共51只,随机分为模型组、丹蛭降糖胶囊高、低剂量组及吡格列酮组。灌胃给药,连续8周。测定大鼠体质量、血糖、血脂、肾功能;肾组织分别用HE、Masson及PAS染色,观察组织的病理变化。结果:与正常组相比,模型组大鼠体质量下降,血糖、血脂显著升高,肾功能明显损害(P<0.01),肾脏病理病变增加,肾脏病理评分显著升高。与模型组相比,各给药组上述指标均有不同程度降低(P<0.05,P<0.01),大鼠肾脏病理形态学明显改善(P<0.05,P<0.01)。结论:丹蛭降糖胶囊能有效改善大鼠的糖尿病症状,减轻肾脏损害。
Objective: To explore the preventive and therapeutic effects of Danzhijiangtang capsule on diabetic nephropathy in rats and provide experimental evidence for its clinical application. Methods: Eighty male Sprague-Dawley rats were randomly selected as the normal control group. The other rats were given high-fat diet plus streptozotocin (STZ) (intraperitoneal injection of 34mg / kg), and the fasting blood glucose / L At the same time, 51 rats with hyperglycemia were excluded and randomly divided into model group, Danzhitangtang capsule high and low dose group and pioglitazone group. Gavage for 8 weeks. The body weight, blood glucose, blood lipid and renal function of rats were measured. The renal tissues were stained with HE, Masson and PAS respectively to observe the pathological changes. Results: Compared with the normal group, the body weight of model rats decreased, the blood glucose and blood lipids were significantly increased, and the renal function was significantly impaired (P <0.01). The pathological changes of kidney increased and the pathological scores of renal increased significantly. Compared with the model group, the above indexes in each administration group were decreased to some extent (P <0.05, P <0.01), and the pathological morphology of the kidney in the administration group was significantly improved (P <0.05, P <0.01). Conclusion: Danzhi Jiangtang Capsule can effectively improve the symptoms of diabetic rats, reduce renal damage.
