目的 比较跨膜型和分泌型TNF α对中性粒细胞生物学功能影响的异同 ,以探讨两型TNF在炎症中的作用。方法 通过吞噬、呼吸爆发、原位杂交等方法研究跨膜型和分泌型TNF α对中性粒细胞生物学功能的影响。结果 分泌型TNF α可促进中性粒细胞吞噬 (P <0 .0 0 1)、呼吸爆发功能 (P <0 .0 5 )及IL 1βmRNA水平的表达 ,对中性粒细胞产生NO无明显影响 ;跨膜型TNF α对中性粒细胞吞噬、呼吸爆发功能无明显影响 ,对IL 1βmRNA水平的表达仅有微弱的促进作用 ,但可明显促进中性粒细胞产生NO。结论 分泌型TNF α对中性粒细胞有明显的激活作用 ,而跨膜型TNF α则可能对其功能具有调节作用
Objective To compare the effects of transmembrane and secretory forms of TNFα on neutrophil biological functions and to explore the role of TNF in inflammation. Methods The effects of transmembrane and secreted TNFα on the neutrophil biological function were studied by phagocytosis, respiratory burst and in situ hybridization. Results Secretion of TNFα could promote neutrophil phagocytosis (P <0.01), respiratory burst function (P <0.05) and IL 1β mRNA expression, and had no effect on NO production by neutrophils ; Transmembrane TNFα had no significant effect on the phagocytosis of neutrophils and respiratory burst function, but had only weak effect on the expression of IL 1β mRNA, but could significantly promote the production of NO by neutrophils. Conclusions Secreted TNFα can obviously activate neutrophils, whereas transmembrane TNFα may play a regulatory role in its function
