目的:建立以毛细管气相色谱法测定莪术油中β-榄香烯、莪术醇、吉马酮和莪术二酮含量的方法。方法:采用气相色谱方法:HP-5(0.32 mm×30 m)石英弹性毛细管柱,进样口温度250℃,氢火焰检测器(FID)280℃,进样量2μL,不分流进样,柱流量1 mL.min-1;程序升温:柱初始温度为100℃,以4℃.min-1升至200℃。采用内标法,以水杨酸甲酯为内标物。结果:β-榄香烯进样量在0.03~0.49μg内与峰面积比值(β-榄香烯/水杨酸甲酯)呈良好的线性关系Y=5.024X-0.0815,r=0.9992,平均回收率98.6%(RSD为2.0%);莪术醇进样量在0.03~0.52μg内与峰面积比值(莪术醇/水杨酸甲酯)呈良好的线性关系Y=4.383X-0.0274,r=0.9993,平均回收率98.0%(RSD为2.5%);吉马酮进样量在0.14~2.20μg内与峰面积比值(吉马酮/水杨酸甲酯)呈良好的线性关系Y=0.969X-0.012,r=0.9997,平均回收率101.0%(RSD为2.3%);莪术二酮进样量在0.13~2.08μg内与峰面积比值(莪术二酮/水杨酸甲酯)呈良好的线性关系Y=0.9047X+0.0268,r=0.9994,平均回收率99.5%(RSD为2.5%)。结论:不同来源的莪术油中4种成分的含量差异显著,莪术二酮在部分样品中未检出。
Objective: To establish a method for the determination of β-elemene, curcumol, germacrone and zedrone by using capillary gas chromatography. Methods: Gas chromatography was used on a HP-5 (0.32 mm × 30 m) quartz capillary column. The inlet temperature was 250 ℃, the hydrogen flame detector (FID) was 280 ℃ and the injection volume was 2 μL. Flow rate 1 mL.min-1; temperature programmed: The initial column temperature was 100 ° C and was raised to 200 ° C at 4 ° C .min-1. Using internal standard method, methyl salicylate as an internal standard. Results: There was a good linear relationship between β-elemene and peak area ratio (β-elemene / methyl salicylate) in the range of 0.03-0.49 g Y = 5.024X-0.0815, r = 0.9992 The recovery rate was 98.6% (RSD was 2.0%). There was a good linear relationship between the concentration of curcumol and the peak area ratio (Curcumol / methyl salicylate) in the range of 0.03-0.52 μg (Y = 4.383X-0.0274, r = 0.9993, the average recovery rate was 98.0% (RSD was 2.5%). There was a good linear relationship between the peak area ratio and the peak area ratio of gemfurone (YM: 0.969X) in the range of 0.14 ~ 2.20μg -0.012, r = 0.9997, the average recovery was 101.0% (RSD was 2.3%). Curcuma dione showed good linearity with the peak area ratio (Curdione / methyl salicylate) in the range of 0.13 ~ 2.08μg Relationship Y = 0.9047X + 0.0268, r = 0.9994, average recovery 99.5% (RSD 2.5%). Conclusion: There are significant differences in the contents of the four components among the different origins of Zedoary turmeric oil. Curdione is not detected in some samples.
