脉塞通是三七的有效成分。为进一步阐明其活血化瘀作用的机理,本实验研究了脉塞通对红细胞膜流动性的影响。实验方法是用DPH作为荧光探针,测定人红细胞膜偏振度。结果表明,当脉塞通的最终浓度为80μg/ml、37℃孵育60min时,能使完整红细胞膜的流动性增加。对照组的荧光偏振度(P)为0.279±0.031,微粘度(η)为3.088±0.326。脉塞通组的P为0.176±0.026,η为1.275±0.306。脉塞通组的P和η与对照组相比较,P值均小于0.001。溶液中脉塞通的浓度大于200μg/ml时,有轻度的溶血作用。脉塞通对红细胞膜流动性的影响是可逆的。
Masaitong is the active ingredient of Panax. In order to further elucidate the mechanism of the effect of promoting blood circulation and removing blood stasis, the effect of gerotonin on the erythrocyte membrane fluidity was studied in this experiment. The experimental method is to use DPH as a fluorescent probe to determine the degree of polarization of human erythrocyte membranes. The results showed that when the final concentration of sarcosine was 80 μg/ml and incubated at 37°C for 60 min, the fluidity of intact erythrocyte membrane was increased. The degree of fluorescence polarization (P) of the control group was 0.279±0.031, and the microviscosity (η) was 3.088±0.326. The P group was 0.176±0.026 and η was 1.275±0.306. Compared with the control group, the P and η of the Masai group were all less than 0.001. When the concentration of nervosa in the solution is greater than 200 μg/ml, mild hemolysis occurs. The effect of gerotonin on the erythrocyte membrane fluidity is reversible.
