目的:研究远志皂苷(tenuifolic saponin,TS)对血压的影响。方法:大鼠麻醉后左颈总动脉记录平均动脉压(MAP)。尾袖法测定清醒大鼠和肾性高血压大鼠(RVHR)收缩压。结果:TS 2,4,8mg·kg~(-1)iv,20和40mg·kg~(-1)ig分别使MAP降低31%,37%,50%,21%和31%;40mg·kg~(-1)ig使清醒大鼠和RVHR的血压分别于给药后60和90min降低38.0%和26.8%,并至少维持2和3h。Dip或切除双侧颈迷走神经合并Atr iv均不影响TS的作用;TS不改变Epi或结扎右颈总动脉所引起的升压反射。结论:TS有降压作用,此作用与迷走神经兴奋,神经节阻断,以及外周α-肾上腺能,M-胆碱能和H_1受体无关。
Objective: To study the effect of tenuifolic saponin (TS) on blood pressure. METHODS: Mean arterial pressure (MAP) was recorded in the left common carotid artery after anesthesia in rats. Caudal-sleeve method was used to determine systolic blood pressure in conscious rats and renal hypertensive rats (RVHR). Results: TS 2, 4 and 8 mg·kg -1 iv, 20 and 40 mg·kg -1 ig reduced MAP by 31%, 37%, 50%, 21% and 31%, respectively; 40 mg·kg ~(-1)ig decreased the blood pressure of conscious rats and RVHR by 38.0% and 26.8% at 60 and 90 min after administration, respectively, and maintained at least 2 and 3 h. Dip or resection of bilateral cervical vagus nerves combined with Atr iv did not affect the effect of TS; TS did not change the boost reflex induced by Epi or ligation of the right common carotid artery. CONCLUSIONS: TS has antihypertensive effects. This effect is not related to vagal excitation, ganglion block, and peripheral α-adrenergic, M-cholinergic, and H_1 receptors.
