目的探讨杜仲提取物对异质性及多因性阿尔茨海默病模型(H/MAD)大鼠空间学习记忆的影响。方法先利用腹腔注射半乳糖促衰老,再反复结扎双侧颈总动脉致脑缺血,最后大鼠侧脑室一次性注射Aβ1-40制备H/MAD模型。将72只健康SD雄性大鼠随机分为6组:空白组、假损伤组、H/MAD模型组、杜仲60%乙醇提取物高剂量组、杜仲60%乙醇提取物中剂量组、安理申组,各给药组于术后3天开始灌胃给药6天。采用Morris水迷宫系统测试各组大鼠的空间学习记忆能力,并测定大鼠血清中T-AOC和TCh E的含量。结果杜仲60%乙醇提取物各组和安理申组均可增强模型大鼠的空间学习记忆能力,能使模型大鼠血清中T-AOC的活力增强,降低TCh E含量,尤以杜仲60%乙醇提取物中剂量组效果最佳。结论杜仲60%乙醇提取物对异质性及多因性阿尔茨海默病模型大鼠的空间学习记忆有明显的改善作用,其机制可能与增强T-AOC的活力和降低TCh E含量有关。
Objective To investigate the effects of Eucommia ulmoides extract on spatial learning and memory in heterogeneous and multiple Alzheimer’s disease (H / MAD) rats. Results Eucommia 60% ethanol extract and allixin group can enhance the spatial learning and memory of the model rats, which can increase the T-AOC activity and reduce the content of TCh E in the serum of the model rats, especially in Eucommia ulmoides 60% ethanol Extract in the dose group best. Conclusion The extract of Eucommia ulmoides 60% ethanol can significantly improve the spatial learning and memory of heterogeneous and multi-factor Alzheimer’s disease rats. The mechanism may be related to the enhancement of T-AOC activity and the reduction of TCh E content.
