目的:分离及鉴定来自于斑马鱼肠道中的酵母菌。方法:采用马铃薯葡萄糖培养基(PDA),从斑马鱼肠道中分离到1株酵母菌,编号为ZF-2,进行形态学观察、生理特征、PCR扩增26S rDNA D1/D2区以及基因测序分析,并构建系统发育进化树。结果:ZF-2分离株为粉红色菌落、菌体呈椭圆形,芽殖;可以同化利用葡萄糖、乳糖、蔗糖等糖类;PCR扩增获得26S rDNA D1/D2区序列长度为642bp(HQ323251),序列比对分析表明与斯鲁菲亚红酵母(Rhodotorula slooffiae)相似性在99%~100%之间,构建的系统发育进化树显示菌株ZF-2与R.slooffiae(EU583485)关系最近,且位于同一分支。结论:首次从斑马鱼肠道中分离到斯鲁菲亚红酵母。
OBJECTIVE: To isolate and identify yeast from the intestinal tract of zebrafish. Methods: One strain of yeast was isolated from zebrafish intestine by using potato dextrose culture medium (PDA), and named ZF-2. Morphological observation, physiological characteristics, PCR amplification of 26S rDNA D1 / D2 region and sequencing analysis , And construct phylogenetic tree. Results: The ZF-2 isolates were pink colonies with oval shape and budding. Assimilable carbohydrates such as glucose, lactose and sucrose were obtained. The 26S rDNA D1 / D2 region was 642bp in length (HQ323251) , And the sequence alignment analysis showed that the similarity to Rhodotorula slooffiae was between 99% and 100%. The phylogenetic tree showed that strain ZF-2 had the closest relationship with R.slooffiae (EU583485) Located on the same branch. Conclusion: Rhizopus sylvestris was isolated from zebrafish intestine for the first time.
