目的:通过蛋白质组技术阐明羊布鲁氏菌外膜蛋白的主要组成。方法:提取并纯化羊布鲁氏菌的外膜蛋白,应用双向电泳技术进行分离,选取双向电泳图上主要的蛋白点进行质谱分析,同时主要蛋白质点的肽指纹图谱利用Mascot在NCBInr蛋白数据库中进行检索。结果:共鉴定到67个蛋白点,主要外膜蛋白为Omp25和Omp31,其他外膜蛋白如Imp、Frp、GroEL等,功能涉及物质运输、能量代谢、应激等。结论:对主要外膜蛋白Omp25和Omp31的识别与分析不仅有助于对布鲁氏菌致病机制的研究,而且为布鲁氏菌病的疫苗研制提供靶标蛋白。
OBJECTIVE: To elucidate the major components of the outer membrane protein of Brucella brucei by proteomic techniques. Methods: The outer membrane protein of Brucella abortus was extracted and purified. The two proteins were separated by two-dimensional electrophoresis. The major protein spots on the two-dimensional gel electrophoresis were selected for mass spectrometry analysis. At the same time, the peptide fingerprinting of major protein spots was performed using Mascot in the NCBInr protein database Search. Results: A total of 67 protein spots were identified. The main outer membrane proteins were Omp25 and Omp31. Other outer membrane proteins such as Imp, Frp and GroEL were involved in the transport of proteins, energy metabolism and stress. Conclusion: The identification and analysis of the major outer membrane proteins Omp25 and Omp31 not only contribute to the study of the pathogenesis of brucellosis, but also provide the target protein for the development of brucellosis vaccine.
