目的探讨不同浓度异丙肾上腺素在体外对婴儿血管瘤内皮细胞生长的影响及β-肾上腺素受体在婴儿血管瘤发病机制中的作用。方法将增殖期婴儿血管瘤内皮细胞分为空白组(不含药物培养基)及异丙肾上腺素低浓度组(5、10、20mg/L)与高浓度组(50、100、200mg/L)。药物作用1~10d采用MTT法分别测定各组细胞的生长曲线。结果低浓度组24~96h对细胞生长呈现刺激作用;96h时,低浓度组与空白对照组的吸光度值差异有统计学意义(P<0.05)。而高浓度组在24~96h对细胞生长呈现抑制作用,与空白对照相比,高浓度组细胞的吸光度值差异有统计学意义(P<0.05),并且24~96h浓度越高,作用越强。结论β-肾上腺素受体在婴儿血管瘤发病中有重要作用;异丙肾上腺素对于体外培养的增殖期血管瘤内皮细胞的刺激作用与浓度相关。
Objective To investigate the effect of different concentrations of isoproterenol on the growth of infantile hemangiomas in vitro and the role of β-adrenergic receptor in the pathogenesis of infantile hemangiomas. Methods The proliferation of infantile hemangiomas endothelial cells were divided into blank group (without drug medium) and isoproterenol low concentration group (5,10,20 mg / L) and high concentration group (50,100,200 mg / L) . Drug effect 1 ~ 10d MTT assay were used to determine the growth curve of each group of cells. Results The cell growth was stimulated in low concentration group (24-96h). At 96h, the difference of absorbance between low concentration group and blank control group was statistically significant (P <0.05). However, the cells in high concentration group showed an inhibitory effect on cell growth at 24-96h. The absorbance values of cells in high concentration group were significantly different from those in blank control group (P <0.05), and the higher the concentration was, the stronger the effect was . Conclusion β-adrenergic receptor plays an important role in the pathogenesis of infantile hemangiomas. Isoproterenol is related to the concentration-dependent stimulating effect on endothelial cells of proliferating hemangioma cultured in vitro.
