目的:探讨邻苯二甲酸二丁酯(DBP)孕期暴露对仔鼠阴茎组织乳酸脱氢酶B(L-lactate dehydrogenase B,LDH-B)的影响,以进一步探讨LDH-B在DBP导致的尿道下裂中的作用机制。方法:将20只受孕大鼠随机分成2组,实验组10只,对照组10只。在大鼠怀孕第14~18天(GD14~18)2组分别给予灌胃,实验组给予DBP每天800mg/kg染毒孕鼠,对照组给予大豆油每天5ml,妊娠19天(GD19)分别取出仔鼠阴茎组织提取总蛋白,通过Western-blot及免疫组化检测LDH-B的表达。结果:实验组及对照组阴茎组织中均有LDH-B的表达,实验组的相对表达量为0.078±0.02(n=10),对照组的相对表达量为0.137±0.03(n=10),实验组中表达明显减少(P<0.05)。LDH-B主要定位于大鼠阴茎组织细胞的胞浆中。结论:孕期大鼠暴露邻苯二甲酸二丁酯后仔鼠阴茎组织中LDH-B表达明显减少,可能与DBP造成机体缺氧有关。
OBJECTIVE: To investigate the effect of DBP exposure during pregnancy on L-lactate dehydrogenase B (LDH-B) in the penile tissue of pups to explore the role of LDH-B in DBP-induced urethra The mechanism of under-cracking. Methods: Twenty pregnant rats were randomly divided into two groups: experimental group (n = 10) and control group (n = 10). The rats in the experimental group were given gavage orally at the 14th to the 18th day of gestation (GD14 ~ 18), and the pregnant rats in the experimental group were administered with DBP 800mg / kg every day. The control group was given 5ml of soybean oil daily for 19 days (GD19) The total protein was extracted from the penile tissue of pups, and the expression of LDH-B was detected by Western-blot and immunohistochemistry. Results: The expression of LDH-B in experimental group and control group was 0.078 ± 0.02 (n = 10) in experimental group and 0.137 ± 0.03 in control group (n = 10) The expression of the experimental group was significantly reduced (P <0.05). LDH-B is mainly located in the cytoplasm of rat penile tissue cells. CONCLUSION: LDH-B expression in the penile tissue of the offspring of pregnant rats exposed to dibutyl phthalate decreased significantly, which may be related to the hypoxia caused by DBP.
