目的观察不同浓度血管紧张奏Ⅱ (Ang Ⅱ )对大鼠肝脏星状细胞 (HSC) [Ca2 +] i 及其增殖的影响 ,以探讨HSC和细胞内 [Ca2 +] i 的相关性。 方法HSC分离培养后 ,采有MTT法和Fura -2 AM荧光指示剂负载分别测定HSC增殖情况及细胞内 [Ca2 +] i。 结果 10 - 9~ 10 - 5mol LAngⅡ能显著增加HSC内游离 [Ca2 +] i(P <0 .0 1) ,且呈剂量依赖关系 ;10 - 9~ 10 - 6mol LAngⅡ均能促进HSC增殖 ( P <0 .0 5 ) ,但 10 - 5mol LAngⅡ却对HSC增殖无显著影响 (P >0 .0 5 )。 结论在一定剂量范围内Ang Ⅱ能够剂量依赖性地促进HSC增殖和增加HSC内游离 [Ca2 +] i,二者存在一定相关性。
Objective To observe the effects of different concentrations of angiotensin Ⅱ on [Ca2 +] i and its proliferation in rat hepatic stellate cells (HSC) to explore the correlation between HSC and intracellular [Ca2 +] i. Methods After isolated and cultured by HSC, MTT assay and Fura -2 AM fluorescent indicator were used to measure the proliferation of HSC and intracellular [Ca2 +] i, respectively. Results 10 - 9 ~ 10 - 5 mol LAng Ⅱ could significantly increase the intracellular [Ca2 +] i in HSC (P <0.01) in a dose - dependent manner. 10 - 9 ~ 10 - 6 mol LAng Ⅱ all promoted the proliferation of HSC <0. 05). However, 10 - 5 mol LAng II had no significant effect on HSC proliferation (P> 0.05). Conclusion Ang Ⅱ can promote the proliferation of HSC in a dose-dependent manner and increase the intracellular [Ca2 +] i in HSC in a dose-dependent manner.
