目的建立一种简单、准确、经济、实用的样品前处理方法,进行鱼油中EPA、DHA的气相色谱法测定。方法应用4种不同的方法对鱼油样品进行前处理,其主要区别在于:分别选用不同的溶剂来进行样品的皂化、萃取。然后按照国家食品安全标准方法 GB 28404—2012分别上机测定,并对测定结果进行了比较。结果 4种前处理方法依次由繁到简,如方法 1要充氮回流皂化,并进行甲酯化、萃取等,步骤较为繁琐。4种不同方法的测定结果采用IBM SPSS Statistics22软件单因素方差分析进行数据处理,发现EPA和DHA检测结果差异无统计学意义(P值分别为0.088、0.695)。结论第4种方法以苯-石油醚溶液为溶剂,进行超声萃取。此方法最为简便实用,节省试剂,节约能源,降低成本。可成为测定鱼油制品中EPA、DHA含量的首选前处理方法。
Objective To establish a simple, accurate, economical and practical sample preparation method for the determination of EPA and DHA in fish oil by gas chromatography. Methods Four different methods were used to pretreat the fish oil samples. The main difference is that saponification and extraction of the samples are carried out by using different solvents respectively. Then according to the national food safety standard method GB 28404-2012 were measured on the machine, and the measured results were compared. Results The four kinds of pretreatment methods followed the procedure from simple to simple, such as method 1 to nitrogen reflux saponification, and methyl esterification, extraction, the more complicated steps. The results of four different methods were analyzed by one-way ANOVA with IBM SPSS Statistics22 software. There was no significant difference in the detection results between EPA and DHA (P = 0.088,0.695, respectively). Conclusion The fourth method using benzene - petroleum ether as solvent, ultrasonic extraction. This method is the most simple and practical, saving reagents, saving energy and reducing costs. It can be the first choice for the determination of EPA and DHA in fish oil products.
