目的:研究益肾调肝方在非酒精性脂肪性肝炎大鼠的预防过程中对线粒体的调控作用。方法:用高脂饲料对SD大鼠造模10周,同时用益肾调肝方及易善复进行预防10周,然后检测各组大鼠肝组织线粒体指标,包括线粒体含量、细胞色素C氧化酶活性、线粒体膜电位、线粒体DNA修复酶等,以及肝功能、血脂及病理,并和正常组及模型组进行比较。结果:益肾调肝方使线粒体含量下降的幅度明显减小(P<0.01);避免了细胞色素C氧化酶活性过度下降(P<0.01);可使线粒体膜电位下降幅度减少(P<0.01);使线粒体DNA修复酶水平明显升高(均P<0.01);另外可以显著降低肝酶及血脂水平,减少脂肪沉积及改善肝脏病理。结论:益肾调肝方预防非酒精性脂肪性肝炎的过程中,对线粒体有明显影响,对线粒体的调控可能是益肾调肝方预防非酒精性脂肪性肝炎的作用靶点之一。
Objective: To study the regulatory effect of Yishen Tiaogan Fang on mitochondria in the prevention of non-alcoholic steatohepatitis in rats. METHODS: Sprague-Dawley rats were modeled with high-fat diet for 10 weeks, and were treated with Yishen Tiaogan Fang and Yi Shanfu for 10 weeks. Then the liver tissue mitochondrial markers including mitochondrial content and cytochrome C oxidase were measured in each group. Activity, mitochondrial membrane potential, mitochondrial DNA repair enzyme, etc., as well as liver function, lipids, and pathology were compared with normal and model groups. Results: Yishen Tiaogan Fang decreased the content of mitochondria significantly (P<0.01), avoided the excessive decrease of cytochrome C oxidase activity (P<0.01), and decreased the decrease of mitochondrial membrane potential (P<0.01). ); The mitochondrial DNA repair enzyme levels increased significantly (all P <0.01); In addition can significantly reduce liver enzymes and blood lipid levels, reduce fat deposition and improve liver pathology. Conclusion: Yishen Tiaogan Recipe has a significant effect on mitochondria in the prevention of non-alcoholic steatohepatitis, and the regulation of mitochondria may be one of the targets of Yishen Tiaogan Fang in preventing non-alcoholic steatohepatitis.
