本文采用体外简易两步灌流法和激素限定RPMI1640培养液对树鼯肝细胞进行分离与培养。结果显示:分离所获肝细胞总数约为1.8×108个,细胞存活率达94%,肝细胞贴壁伸展能力强,培养时间较长,2周内保持良好的形态特征。认为分离培养的树鼯肝细胞可用作肝病研究的体外细胞模型,有助于人肝炎病毒感染的进一步研究。
In this paper, a simple two-step in vitro perfusion method and hormone-limited RPMI1640 medium for isolation and culture of tree shrews liver cells. The results showed that the total number of isolated hepatocytes was about 1.8 × 108, the cell survival rate was 94%, the ability of hepatocyte adhering to the wall was strong, the culture time was longer, and the morphological characteristics were maintained within 2 weeks. It is considered that isolated and cultured tree shrew hepatocytes can be used as an in vitro cell model for liver disease research and contribute to the further study of human hepatitis virus infection.
