采用UHPLC-QTOF-MS技术,对吴茱萸水提物的主要成分及给药后大鼠体内的原形成分及其代谢产物进行定性分析。通过分析各成分的保留时间、精确分子质量、一级、二级质谱信息并与相应的对照品比对,从吴茱萸水提物中鉴定了27个成分,其中25个成分经对照品确证;从灌胃给予吴茱萸水提物的大鼠血浆、尿液和粪便中鉴定、推测出16个原形成分和35个代谢产物,16个原形成分均经对照品确证。吴茱萸水提物的主要成分有酚酸、黄酮苷、柠檬苦素和生物碱。生物碱、黄酮和酚酸存在于血浆、尿液和粪便中,柠檬苦素主要存在于尿液和粪便中。这些成分在大鼠体内代谢的主要途径有羟基化、氢化、甲基化、硫酸化、葡糖醛酸化等。该研究基本明确了吴茱萸水提物中各类成分在大鼠体内的主要存在形式,为吴茱萸体内药效物质的阐明提供了参考。
Using UHPLC-QTOF-MS technology, the main constituents of Evodia rutaecarpa water extract and qualitative analysis of the prototype components and their metabolites in rats after administration. 27 components were identified from Evodia rutaecarpa water extracts by analyzing the retention time, accurate molecular mass, primary and secondary MS information of each component, and comparing with the corresponding reference substance. Among them, 25 components were confirmed by reference substance; Fructus Evodia given aqueous extract of rat plasma, urine and stool identification, estimated 16 prototype components and 35 metabolites, 16 prototype ingredients were confirmed by the reference substance. The main components of Evodia water extract are phenolic acids, flavonoid glycosides, limonoids and alkaloids. Alkaloids, flavonoids and phenolic acids are present in plasma, urine and excrement, and limonoids are predominantly found in urine and excrement. The main routes of metabolism of these components in rats are hydroxylation, hydrogenation, methylation, sulfation, glucuronidation and so on. This study basically clarified the main forms of various components of Evodia rutaecarpa water extract in rats and provided a reference for the elucidation of pharmacodynamics substances in Evodia rutaecarpa.
