以油桐无菌苗叶柄为外植体,研究植物生长调节剂对其离体培养及植株再生的影响。结果表明:叶柄直接诱导不定芽的最佳培养基1/2MS+3.0 mg·L-1 6-BA+0.05 mg·L-1 IAA,诱导率达91.67%;最佳继代增殖培养基1/2MS+3.0 mg·L-16-BA+0.05 mg·L-1 IBA+1.0 mg·L-1 GA3,增殖系数可达4.89;最佳生根培养基为1/2MS+0.05 mg·L-1 IBA,生根率96.18%。炼苗移栽到泥炭土:珍珠岩:黄土=2:1:1的基质中,成活率达93.55%以上。
The effect of plant growth regulators on in vitro culture and plant regeneration was studied using the leaves of A. tungi as explants. The results showed that the induction rate of adventitious shoots was 1 / 2MS + 3.0 mg · L-1 6-BA + 0.05 mg · L-1 IAA, and the induction rate was 91.67%. The optimal subculture proliferation medium 1 / 2MS + 3.0 mg · L-16-BA + 0.05 mg · L-1 IBA + 1.0 mg · L-1 GA3, the proliferation coefficient was 4.89. The best rooting medium was 1/2 MS + 0.05 mg · L -1 IBA , Rooting rate of 96.18%. The seedling transplanting to peat soil: perlite: loess = 2: 1: 1 matrix, the survival rate of 93.55% or more.
