将金黄色葡萄球茵A蛋白纯品和含A蛋白的金黄色葡萄球菌以不同剂量、不同途径注入615近交系小鼠体内。采用微核测定法检测小鼠外周血淋巴细胞微核和双核。表明SpA纯品和SpA菌体对小鼠淋巴细胞染色体均有损伤作用。SpA纯品0.5mg/只腹腔注射诱导小鼠外周血淋巴细胞微核率达7‰,双核率达30.45‰,对淋巴细胞染色体的损伤程度最重。各组的SpA茵体诱导小鼠外周血淋巴细胞的双核率明显高于微核率。由此提出把双核测定也作为检测致突变物致突变作用的一种方法。
The pure A. aureus and A protein-containing Staphylococcus aureus were injected into 615 inbred mice at different doses and in different ways. Micronucleus assay was used to detect micronuclei and binucleated lymphocytes in peripheral blood of mice. It was shown that SpA pure product and SpA bacteria had damage to chromosome of mouse lymphocytes. SpA pure product 0.5mg/only intraperitoneal injection induced peripheral blood lymphocyte micronucleus rate of 7 ‰, dual-nuclear rate of 30.45 ‰, the most serious damage to the lymphocyte chromosome. The rate of dual-nuclear lymphocytes in peripheral blood lymphocytes induced by SpA in each group was significantly higher than that in micronucleus. Therefore, the dual-core assay is also proposed as a method for detecting mutagenicity of mutagens.
