百草枯对健康者外周血单个核细胞OPN,NF-KB的影响

目的:探讨百草枯对健康者外周血单个核细胞(peripheral blood mononuclear cell PBMC)骨桥蛋白(osteopontin OPN),核转录因子-KB(Nuclear factor-kB,NF-KB)的影响。方法:健康体检者6例,取外周静脉血分离获得PBMC,以5×105个细胞/孔种入96孔细胞培养板,分3组培养12小时。A1组:正常PBMC; A2组:正常PBMC+50ng/ml百草枯;A3组:正常PBMC+100n g/m l百草枯。观察培养前后细胞形态变化,收集各组细胞培养上清检测骨桥蛋白水平,收获细胞后检测胞内N F-K B。结果:各组细胞培养前后细胞形态、数量无明显变化;从A1组到A3组,细胞培养上清骨桥蛋白水平及胞内N F-K B含量呈逐组增高。结论:百草枯影响细胞内NF-KB,导致骨桥蛋白的大量分泌,可能是其引起肺组织损伤的原因。“,”Objective To investigate the synergetic effect of paraquet on osteopontin(OPN), Nuclear factor-kB from peripheral blood mononuclear cell(PBMC) in healthy volunteers.Methods Peripheral blood mononuclear cells from 6 healthy volunteers were obtained after centrifugalization,5×105PBMCs/well seeded in 96 wells cell culture plate were divided into 3 groups for cultivating 12 hours.A1 group;normal PBMC;A2 group:normal PBMC+50ng/ml paraquet;A3 group: normal PBMC+100ng/ml paraquet. Cell morphologic changes before and after culture were observed, OPN of culture supernatant were detected by ELISA, NF-KB within cells were detected by western blot.Results Cell appearance and amount before and after 12 hours culture didn’t show marked change in three groups;There were obvious difference about levels of OPN within three groups;So was level of NF-KB .Conclusions Paraquet significantly increased OPN of production of PBMC,raised levels of NF-KB within PBMC,maybe gradually induce lung organization damage.
