目的:探讨人类端粒酶逆转录酶(humantelomerasereversetranscriptase,hTERT)基因的反义寡核苷酸(antisenseoligodeoxynucleotide,ASODN)对K562细胞端粒酶活性的影响。方法:采用多聚酶链反应-酶联免疫测定(PCR-ELISA)方法检测人工合成的反义脱氧寡核苷酸处理K562细胞前后端粒酶活性的改变,以逆转录PCR(RT-PCR)分析hTERT基因mRNA表达水平。结果:hTERT基因的ASODN作用于K562细胞后,hTERTmRNA表达水平显著下调,其端粒酶活性明显受到抑制。结论:hTERT基因的ASODN可以在翻译水平显著抑制K562细胞的端粒酶活性,hTERT基因与端粒酶的活性密切相关。
Objective: To investigate the effect of antisense oligodeoxynucleotide (ASODN) on the telomerase activity of K562 cells induced by human telomerase reverse transcriptase (hTERT) gene. Methods: The changes of telomerase activity in K562 cells treated with antisense oligonucleotide were detected by polymerase chain reaction-enzyme-linked immunosorbent assay (PCR-ELISA), and hTERT Gene mRNA expression level. Results: The hTERT mRNA expression was significantly down-regulated by ASODN of hTERT gene in K562 cells, and the telomerase activity was significantly inhibited. CONCLUSION: ASODN of hTERT gene can significantly inhibit the telomerase activity of K562 cells at translation level. The hTERT gene is closely related to the activity of telomerase.
