目的观察钙结合蛋白S100A2在原发性肝癌中的表达及与肿瘤细胞增殖的关系,探讨两者对肝癌患者预后的影响。方法采用免疫组织化学链霉菌抗生物素蛋白-过氧化酶(SP)法检测62例肝癌标本中S100A2的表达,同时用计算机图象分析技术对其DJ值(DNA含量指数)进行检测。结果高/中分化组,无门脉/胆管浸润或淋巴结/远处转移的病例中,S100A2阳性表达率分别为77.27%、76.92%、80.00%;DI值分别为1.68±0.20、1.73±0.23、1.76±0.23;但随着肝癌分化程度的降低,出现门脉/胆管浸润或发生淋巴结/远处转移,S100A2阳性表达率降低(50.00%、47.22%、50.00%)(X~2=4.39、5.53、5.07,P均<0.05),而DI值明显升高(1.93±0.29、1.99±0.31、2.01±0.33)(t=3.612、3.794、3.379,P均<0.01)。并且在短/中期生存患者中,S100A2阳性表达率(50.00%)明显低于长期生存者(100%)(X~2=10.05,P<0.01),而DI值(1.88±0.25)却明显高于长期生存者(1.69±0.21)(t=2.606,P<0.05)。S100A2的表达与DI值呈明显负相关(t=3.023,P<0.05)。结论S100A2蛋白表达降低和高的DI值提示肝癌的低分化、高侵袭转移能力及预后不良,两者的联合检测对判断肝癌的恶性程度和预后具有重要的临床意义。
Objective To observe the expression of calcium binding protein S100A2 in primary hepatocellular carcinoma (HCC) and its relationship with the proliferation of tumor cells, and to explore the influence of S100A2 on the prognosis of HCC patients. Methods Immunohistochemical streptavidin-peroxidase (SP) method was used to detect the expression of S100A2 in 62 specimens of hepatocellular carcinoma, and its DJ value (DNA content index) was detected by computer image analysis. Results The positive rates of S100A2 in high / moderately differentiated group, without portal / bile duct invasion or lymph node / distant metastasis were 77.27%, 76.92% and 80.00%, respectively; the DI values were 1.68 ± 0.20 and 1.73 ± 0.23, 1.76 ± 0.23. However, with the decrease of the degree of differentiation of hepatocellular carcinoma, the positive rate of S100A2 decreased (P <0.05), the portal vein / bile duct infiltration or lymph node / distant metastasis decreased (50.00%, 47.22%, 50.00% , 5.07, P <0.05), while the DI value increased significantly (1.93 ± 0.29, 1.99 ± 0.31, 2.01 ± 0.33) (t = 3.612,3.794,3.379, P <0.01). (50.00%) was significantly lower than that of long-term survivors (X ~ 2 = 10.05, P <0.01), while the DI value (1.88 ± 0.25) was significantly higher in short / medium survival patients In long-term survival (1.69 ± 0.21) (t = 2.606, P <0.05). The expression of S100A2 was negatively correlated with DI (t = 3.023, P <0.05). Conclusions The decrease of S100A2 protein expression and high DI value suggest the poorly differentiated, highly invasive and metastatic potential of HCC as well as poor prognosis. The combined detection of S100A2 protein and HIF-1α protein has important clinical significance in judging the malignant degree and prognosis of HCC.
