目的 研究一种神经营养物质CBD和尼莫地平(Nimodipine,Nim)联合应用对培养大鼠皮层神经元损伤的保护作用。方法 分别采用谷氨酸化学或机械划痕创伤性损伤培养大鼠皮层神经元,根据培养细胞台盼蓝活力计数和培养上清乳酸脱氢酶(LDH)检测评估损伤程度。结果 CBD和Nim单独应用时,对谷氨酸和创伤介导的培养皮层神经元损伤有中等程度的保护作用,与单独损伤未加保护性药物组相比较有显著性差异(P<0.01)。CBD和Nim联合会用时,保护作用比单独应用明显增强(P<0.01)。结论 CBD和Nim联合应用有明显的协同作用。
Objective To study the protective effect of a combination of CBD and Nimodipine (Nimodipine, Nim), a neurotrophic substance, on cultured rat cortical neurons. Methods Rat cortical neurons were cultured with traumatic injury induced by chemical or mechanical scratching of glutamate. The degree of injury was evaluated according to the viability count of cultured cells and the culture supernatant lactate dehydrogenase (LDH). Results When CBD and Nim were used alone, there was a moderate degree of protective effect on glutamate-induced and trauma-induced neuronal damage in cultured cortical neurons, which was significantly different from that of the unprotected drug group alone (P <0.01). Compared with single application, the protection effect of CBD and Nim association was significantly enhanced (P <0.01). Conclusion The combination of CBD and Nim has obvious synergistic effect.
