目的探讨低剂量棉酚和甾体激素联合用药对睾丸精母细胞数量、凋亡和支持细胞吞噬作用的影响。方法本实验采用大鼠喂服棉酚与甾体激素联合用药方式[棉酚12.5mg/(kg·d)+去氧孕烯125μg/(kg·d)+炔雌醇25μg/(kg·d)+十一酸睾丸酮100mg/(kg·d)],与单独喂服相同剂量的棉酚或激素的大鼠及喂服载体溶剂的大鼠相对照,用药4、6和8周取睾丸组织,应用体视学、原位缺口末端标记(TUNEL)和油红O染色的方法,观察精母细胞和球形精子细胞数量、凋亡和支持细胞吞噬作用。结果联合用药中,甾体激素成分可明显减少精母细胞和球形精子细胞的数量,精母细胞凋亡增加并同时伴有生精上皮油红O染色的增强。结论精母细胞凋亡并被支持细胞吞噬是联合用药抗生育机制之一。
Objective To investigate the effects of combination of low dose gossypol and steroid hormone on the number of spermatocyte, apoptosis and phagocytosis of supporting cells in testis. Methods In this experiment, rats were fed with gossypol and steroid hormone combined with gossypol (12.5mg / (kg · d) + desogestrel 125μg / (kg · d) + ethinyl estradiol 25μg / (kg · d ) + Testosterone undecanoate (100 mg / (kg · d)] compared with rats fed the same dose of gossypol or hormone and vehicle-fed vehicle alone. Testis tissues were taken at 4, 6 and 8 weeks The numbers of spermatocytes and spheroids, apoptosis and phagocytosis of supporting cells were observed by stereological method, TUNEL and oil red O staining. Results In the combination therapy, steroid hormones significantly reduced the number of spermatocytes and spherical spermatids, increased spermatocyte apoptosis accompanied by enhanced spermatogenic epithelial oil red O staining. Conclusions Apoptosis of spermatocytes and their engulfment by supporting cells are one of the mechanisms of combined anti-fertility therapy.
