目的研究白花蛇舌草乙酸乙酯提取液对多药耐药白血病细胞HL-60/ADR的抑制作用及其机制。方法采用MTT比色试验观察白花蛇舌草乙酸乙酯提取液对多药耐药细胞HL-60/ADR的抑制作用。采用光镜、电镜技术观察细胞形态结构的改变,利用流式细胞术检测细胞凋亡率。结果①白花蛇舌草乙酸乙酯提取液能明显抑制HL-60/ADR细胞的生长,药物浓度相当于生药41.67mg/ml时,即具有显著的抑制作用,抑制率呈剂量和时间依赖性,半数抑制浓度(IC50)相当于生药70.41mg/ml。②形态学改变,呈典型的凋亡特征。③白花蛇舌草乙酸乙酯提取液能诱导多药耐药细胞HL-60/ADR凋亡,凋亡率也呈时间和剂量的依赖性。结论白花蛇舌草对多药耐药细胞HL-60/ADR的生长具有极强的抑制作用,诱导其凋亡是其机制之一。
Objective To study the inhibitory effect of ethyl acetate extracts of Hedyotis diffusa Willd on the multidrug-resistant leukemia cells HL-60 / ADR and its mechanism. Methods MTT colorimetric assay was used to observe the inhibitory effect of ethyl acetate extract of Hedyotis diffusa Willd on the multidrug resistance cell line HL-60 / ADR. The morphological changes of cells were observed by light microscope and electron microscope. The apoptosis rate was detected by flow cytometry. Results The extract of Hedyotis diffusa Willd. Could significantly inhibit the growth of HL-60 / ADR cells. When the drug concentration was equivalent to 41.67 mg / ml crude drug, the extract had significant inhibitory effect in dose and time dependent manner, The median inhibitory concentration (IC50) is equivalent to 70.41 mg / ml crude drug. ② morphological changes, typical apoptotic characteristics. ③ Hedyotis diffusa extract ethyl acetate can induce multi-drug resistant cell HL-60 / ADR apoptosis, apoptosis rate also showed time and dose-dependent. Conclusion Hedyotis diffusa has a strong inhibitory effect on the growth of multidrug-resistant cells HL-60 / ADR. Inducing apoptosis is one of the mechanisms.
