目的研究右归丸对“劳倦过度、房室不节”肾阳虚小鼠模型脑基因的影响,以期从基因水平上研究其药物作用机理。方法对照组、模型组和治疗组雄性小鼠各为10只、15只和10只。对照组和模型组每日每只灌胃蒸馏水0.5ml,治疗组每日每只予右归丸混悬液0.5ml,模型组和治疗组雄雌鼠比例1∶6同笼喂养且雄鼠每日游泳30~40分钟共4周,以诱导产生“劳倦过度、房室不节”肾阳虚证。用36K鼠脑芯片检测3组鼠脑基因,并各以二者荧光信号相对强度比值≥2和≤0.5筛选差异显著基因。结果模型组/对照组共筛选出186个差异表达基因,其中上调基因150个,下调基因36个。治疗组/模型组筛选出155个差异表达基因,其中上调基因64个,下调基因91个。模型组/对照组上调而治疗组/模型组下调的基因有32个,主要是炎症/免疫、神经传递/信号转导等基因。模型组/对照组下调而治疗组/模型组上调的基因有12个,主要是与黑色素、孤儿核受体相关的基因。结论右归丸可使肾阳虚小鼠模型显著下调的黑色素显著上调,改善与孤儿核受体有关激素的作用并促进细胞增殖。
Objective To study the effect of Yougui Pills on brain gene expression in mouse model of kidney fatigue with overwork fatigue and atrioventricular dysfunction, so as to study its mechanism of action from the genetic level. Methods There were 10, 15 and 10 male mice in the control, model and treatment groups. The control group and the model group were treated with 0.5 ml distilled water per day, and the treatment group received 0.5 ml of the right-give pill suspension each day. The ratio of the male and female rats in the model group and the treatment group was the same cage fed 1:6 and the male rats were daily. Swim 30-40 minutes for a total of 4 weeks to induce “overwork, atrioventricular” and “deficiency of kidney yang”. Three groups of rat brain genes were detected using a 36K mouse brain chip, and the differentially significant genes were screened by the ratio of the relative intensity of the fluorescence signals of the two groups ≥ 2 and ≤ 0.5. Results A total of 186 differentially expressed genes were screened out in the model/control group, of which 150 were up-regulated and 36 were down-regulated. 155 differentially expressed genes were screened out in the treatment/model group, of which 64 were up-regulated and 91 were down-regulated. In the model group/control group, there were 32 genes down-regulated in the treatment group/model group, mainly inflammatory/immune, neurotransmission/signal transduction and other genes. There were 12 genes up-regulated in the model group/control group and up-regulated in the treatment group/model group, mainly related to melanin and orphan nuclear receptors. Conclusion Yougui pill can significantly up-regulate melanin in kidney-yang deficiency mice model, improve the role of orphan nuclear receptor-related hormones and promote cell proliferation.
