目的:探讨荧光偏振免疫法测定甲氨蝶呤血药浓度时样品的保存时间、保存条件对测定结果的影响。方法:取接受大剂量甲氨蝶呤化疗后的小儿急性淋巴细胞白血病患者的静脉血,用荧光偏振免疫法分别测定冰箱冷藏、室温避光、室温不避光保存1,2d后的血药浓度,以考察保存时间、温度、光线对测定结果的影响。结果:冰箱冷藏、室温避光和室温不避光保存1,2d后测定结果均与保存前的测定值无统计学差异。结论:甲氨蝶呤血样在室温不避光条件下可稳定保存2d。
OBJECTIVE: To investigate the influence of the storage time and the storage conditions of the sample on the determination of the concentration of methotrexate by fluorescence polarization immunoassay. Methods: The venous blood of children with acute lymphoblastic leukemia after receiving high-dose methotrexate chemotherapy was measured by fluorescence polarization immunoassay, refrigerator was stored at room temperature, dark, at room temperature after dark preservation for 1, 2 days, the plasma concentration , To examine the preservation time, temperature, light on the measurement results. Results: There was no significant difference between the results of refrigerated refrigerator, dark at room temperature and room temperature without dark after 1 and 2 days of storage. Conclusion: Methotrexate blood samples can be stably stored for 2 days at room temperature without light.
