根据杂交水稻种子稻壳来自母本及SSR分子标记的共显性特点,提出了一种基于杂交水稻种子鉴别其亲本的分子检测技术。首先提取杂交水稻种子稻壳的DNA,获得母本的DNA指纹图谱,再结合杂种带型,推导出父本带型。这样只要建立已有亲本的DNA指纹图谱库,通过比对,就可以鉴别杂交水稻的亲本是已登记亲本还是新创亲本。以金优207及其亲本作为实验材料,选用5对SSR引物验证了上述检测技术。
According to the co-dominant characteristic of hybrid rice seed hulls derived from the female parent and SSR markers, a molecular detection technique based on hybrid rice seeds for identifying their parents was proposed. Firstly, the DNA of the rice husk of hybrid rice was extracted to obtain the DNA fingerprints of the female parent. In this way, as long as a DNA fingerprinting database of existing parents is established, the parents of the hybrid rice can be identified as registered parents or new parents by comparison. Taking Jinyou 207 and its parents as experimental materials, five pairs of SSR primers were selected to verify the above detection techniques.
