目的:探讨金匮肾气丸对庆大霉素造成药物性耳聋豚鼠耳蜗血管纹细胞凋亡调控基因表达的影响。方法:豚鼠腹腔注射庆大霉素100mg·kg~(-1)·d~(-1)制作耳聋动物模型,治疗组在注射庆大霉素后灌服金匮肾气丸药剂,实验结束后豚鼠断头处死,取耳蜗标本。观察指标:免疫组化B细胞淋巴瘤/白血病-2(Bcl-2)及Bcl-2相关蛋白(Bax)的表达定量检测分析。结果:Bax蛋白表达:与空白组比较,模型组Bax蛋白表达增多,差异有非常显著性意义(P<0.01);与治疗组比较,模型组Bax蛋白表达增多,差异有显著性意义(P<0.05);治疗组与空白组Bax蛋白表达比较,差异无显著性意义(P>0.05)。Bcl-2蛋白表达:与空白组、治疗组比较,模型组Bcl-2蛋白表达明显减少,差异有非常显著性意义(P<0.01);治疗组与空白组Bcl-2蛋白表达比较,差异无显著性意义(P>0.05)。结论:金匮肾气丸可以降低血管纹侧壁Bax蛋白表达,调节Bcl-2蛋白表达,从而抑制血管纹细胞凋亡,改善血液循环,达到防治耳聋的作用。
Objective: To investigate the effect of Jinkui Shenqi Pills on the expression of apoptosis-regulating genes in guinea pig cochlear stria vascularis induced by gentamicin in drug-induced deafness. METHODS: Guinea pigs were injected intraperitoneally with gentamicin 100 mg·kg -1 ·d -1 to make animal models of deafness. The treatment group was given Jintanshenqi Pills after gentamycin injection, after the experiment was completed. The guinea pig was killed by decapitation and the cochlea specimens were taken. OBJECTIVES: Quantitative analysis of expression of immunohistochemical B-cell lymphoma/leukemia-2 (Bcl-2) and Bcl-2 associated protein (Bax). Results: Bax protein expression: compared with the blank group, the expression of Bax protein in the model group increased, the difference was significant (P <0.01); compared with the treatment group, the expression of Bax protein in the model group increased, the difference was significant (P < 0.05); There was no significant difference in the expression of Bax protein between the treatment group and the blank group (P>0.05). Bcl-2 protein expression: Compared with the blank group and the treatment group, the Bcl-2 protein expression in the model group was significantly reduced (P<0.01); the expression of Bcl-2 protein in the treatment group and the blank group was compared, the difference was not Significant significance (P>0.05). Conclusion: Jinkui Shenqi Pill can reduce the expression of Bax protein in the stria vascularis, regulate the expression of Bcl-2 protein, inhibit the apoptosis of stria vascularis cells, improve the blood circulation, and achieve the effect of preventing and treating deafness.
