将尖海龙用水、磷酸缓冲液、40%乙醇、水煎4种不同方法进行提取,再将4种不同提取物分别作用于Hela、GLT-82、SK-RB-3细胞系,于加药后1d、2d、3d倒置显微镜观察细胞的形态变化,四种提取物3种肿瘤细胞系均有杀伤,且随加药浓度的增加、时间的延长,杀伤效力增强。加药后3d,用MTT法测定细胞的存活率,几种提取方法结果相差不大,对海龙的有效抗癌成分仍不能进行准确的定位,推测海龙的抗癌成分可能不是大分子.也不是脂溶性物质,可能是水溶性的小分子物质。
Five different methods were used to extract water, phosphate buffer, 40% ethanol and decocted water. Four different extracts were applied to the Hela, GLT-82 and SK-RB-3 cell lines, respectively. The morphological changes of the cells were observed by inverted microscope at 1d, 2d, and 3d. All three tumor cell lines of the four extracts showed killing. With the increase of drug concentration and time, the killing effect was enhanced. After 3 days of dosing, the viability of the cells was measured by MTT assay. The results of several extraction methods were similar, and the effective anti-cancer components of Hailong could not be accurately located. It was inferred that the anti-cancer components of Hailong may not be macromolecules. Fat-soluble substances may be water-soluble small molecules.
