目的:观察仙虎合剂对大鼠实验性脂肪肝的治疗作用。方法:Wistar大白鼠50只,随机分为正常对照组、模型组、中药仙虎合剂高、低剂量组和甲硫氨酸组。除正常对照组外,其余4组均以高脂饮食喂养10周,建立高脂血症性脂肪肝模型后,模型组用生理盐水ig,中药高、低剂量组用仙虎合剂(相当于生药31.5,15.75 g.kg-1)ig,甲硫氨酸组用甲硫氨酸片(0.625 g.kg-1)ig。各组于用药4周后处死取材送检,观察肝体比、血脂、肝脂及肝组织病理改变等。结果:模型组镜下呈重度脂肪肝改变,血清总胆固醇(TC)、甘油三酯(TG)分别为(3.09±0.84),(2.29±0.41)mmol.L-1,肝组织匀浆TC,TG分别为(5.87±0.65),(23.46±3.28)mmol.L-1,与正常对照组比较有显著性差异(P<0.01);仙虎合剂高剂量组镜下接近于正常表现,血清TC,TG分别为(1.68±0.22),(1.73±0.19)mmol.L-1,肝组织匀浆TC,TG分别为(3.42±0.44),(16.47±2.52)mmol.L-1,与模型组比较有显著性差异(P<0.01);仙虎合剂高、低剂量组血清TC(1.68±0.22),(1.72±0.36)mmol.L-1,与甲硫氨酸组(2.14±0.53)mmol.L-1比较有显著性差异(P<0.05);3个治疗组其他各项数据相近,提示仙虎合剂能明显降低脂肪肝大鼠血脂,减轻肝脂变及肝细胞坏死程度。结论:仙虎合剂对高脂饮食诱导的脂肪肝具有确切治疗作用。
Objective: To observe the therapeutic effect of Xianhu mixture on experimental fatty liver in rats. Methods: Fifty Wistar rats were randomly divided into normal control group, model group, Sinus Huatan high and low dose group and methionine group. Except for the normal control group, the other four groups were fed with a high-fat diet for 10 weeks. After establishing the model of hyperlipidemic fatty liver, the model group was treated with normal saline ig and high and low doses of Chinese traditional medicine (equivalent to crude drug 31.5, 15.75 g.kg-1) ig, methionine group methionine tablets (0.625 g.kg-1) ig. The rats in each group were sacrificed after 4 weeks of treatment for inspection, liver and body ratio, blood lipids, liver fat and pathological changes of liver tissue. Results: The model group showed severe fatty liver changes, the total cholesterol (TC) and triglyceride (TG) were (3.09 ± 0.84), (2.29 ± 0.41) mmol.L- TG were (5.87 ± 0.65) and (23.46 ± 3.28) mmol.L-1, respectively, which were significantly different from the normal control group (P <0.01) , TG (1.68 ± 0.22) and (1.73 ± 0.19) mmol.L-1, respectively. The levels of TC and TG in liver homogenate were (3.42 ± 0.44) and (16.47 ± 2.52) mmol.L- (1.68 ± 0.22) and (1.72 ± 0.36) mmol.L-1 in the high and low dose groups of Xianhu Mixture group and 2.14 ± 0.53 mmol / L in the methionine group (P <0.01) (P <0.05). The other data of three treatment groups were similar, which indicated that Xianhu Mixture could obviously reduce the blood lipid, reduce the degree of hepatic steatosis and hepatocellular necrosis in rats with fatty liver. Conclusion: Xianhu Mixture has the exact therapeutic effect on fatty liver induced by high-fat diet.
