目的观察病毒性心肌炎小鼠急性期和慢性期N-cadhein表达变化,并探讨其机制。方法用CVB3反复感染Balb/c小鼠,分别于感染后第10d、5个月不同时间点,利用高频探头行超声检查心功能,取心肌经HE染色观察其病理变化,并用免疫组织化学,Western-blot以及RT-PCR方法对心肌组织N-cadherin表达的变化进行定性和定量分析。结果第10d,心脏急性炎性反应明显,心功能下降,但心肌组织N-cadherin表达与对照组无明显差异。第5月,实验组小鼠心脏出现扩张型心肌病样改变。实验组小鼠心肌组织N-cadherin表达较对照组明显下调。结论心肌组织N-cadherin表达的变化可能是小鼠病毒性心肌炎发展的重要机制之一。
Objective To observe the changes of N-cadherin expression in acute and chronic viral myocarditis in mice and to explore its mechanism. Methods Balb / c mice were repeatedly infected with CVB3. The heart function was examined ultrasonically with high frequency probe at different time points after 10 days and 5 months respectively. Pathological changes of myocardium were observed by HE staining. Immunohistochemistry, Western-blot and RT-PCR methods were used to qualitatively and quantitatively analyze the changes of N-cadherin expression in myocardial tissue. Results On the 10th day, the acute inflammatory reaction of the heart was obvious and the cardiac function was decreased. However, the expression of N-cadherin in myocardial tissue was not significantly different from that in the control group. The fifth month, experimental mice with heart-shaped dilated cardiomyopathy-like changes. The expression of N-cadherin in the experimental group was significantly lower than that in the control group. Conclusion The change of N-cadherin in myocardium may be one of the important mechanisms of the development of viral myocarditis in mice.
