[目的]研究TNF-α对鼻咽癌中c-IAP1和c-IAP2表达水平的影响,探索鼻咽癌抗凋亡的机制。[方法]培养鼻咽癌5-8F细胞,经不同浓度(0、1、5、10、20、30ng/ml)的TNF-α处理细胞16h、24h之后,半定量RT-PCR检测c-IAP1和c-IAP2基因表达水平的变化,用WesternBlot检测c-IAP1和c-IAP2蛋白表达水平的变化。[结果]低浓度TNF-α上调鼻咽癌中c-IAP1表达,而高浓度TNF-α下调c-IAP1表达。随着TNF-α浓度增高,c-IAP2表达量不断上调。[结论]TNF-α通过上调鼻咽癌细胞中c-IAP2的表达而使肿瘤细胞抗凋亡,可能成为抗癌靶点。
[Objective] To investigate the effect of TNF-α on the expression of c-IAP1 and c-IAP2 in nasopharyngeal carcinoma and to explore the mechanism of anti-apoptosis of nasopharyngeal carcinoma. [Methods] The nasopharyngeal carcinoma 5-8F cells were cultured and treated with TNF-α at different concentrations (0, 1, 5, 10, 20 and 30 ng / ml) for 16 h. After 24h, the expression of c-IAP1 And c-IAP2 gene expression levels were detected by Western Blot c-IAP1 and c-IAP2 protein expression levels. [Results] The low concentration of TNF-α up-regulated the expression of c-IAP1 in nasopharyngeal carcinoma and the high concentration of TNF-α down-regulated the expression of c-IAP1. With the increase of TNF-α concentration, the expression of c-IAP2 continuously increased. [Conclusion] TNF-α may be an anti-cancer target by up-regulating the expression of c-IAP2 in nasopharyngeal carcinoma cells.
