目的研究利用骨髓库HLA资料建立高匹配诱导多能干细胞(iPS)库的可行性。方法通过对中华骨髓库湖南分库长沙实验室5 236(人)份HLA分型数据统计分析,找出O型HLA-A、-B、-DRB1三座位纯合子数据,计算以纯合子志愿者为假定iPS供者、5 236名随机志愿者为假定患者的匹配几率;同时计算随机志愿者HLA单体型频率,评估以O型HLA高频单体型纯合子志愿者为假设iPS供者的匹配几率。结果在5 236份骨髓供者资料中共发现O型HLA单体型纯合子14种,以此建库可以分别为49.81%(2 608/5 236)、61.29%(3 209/5 236)、79.51%(4 163/5 236)、75.76%(3 967/5 236)及97.78%(5 120/5 236)的假定患者提供5种匹配水平的iPS供者;共检出17种高频率(HF>1%)的单体型,以这些单体型的O型纯合子建库可以分别为64.06%(3 354/5 236)、75.55%(3 956/5 236)、81.11%(4 247/5 236)、78.76%(4 124/5 236)及98.28%(5 146/5 236)的假定患者提供5种匹配水平的iPS供者。结论利用中华骨髓库现有的HLA资料可以建立数量少、效率高的iPS细胞库。
Objective To study the feasibility of establishing a library of high matching induced pluripotent stem cells (iPS) using HLA data from bone marrow. Methods According to the statistical analysis of 5 236 (human) HLA typing data from Changsha Laboratory of Hunan Bone Marrow Bank in Changsha Branch, the data of homozygous O-type HLA-A, -B, -DRB1 loci were obtained. The homozygous volunteers Suppose iPS donors, 5 236 random volunteers for the assumed probability of matching patients; at the same time calculate the frequency of HLA haplotypes in random volunteers to assess the type O HLA haplotype haplotype homozygous volunteers for the hypothesized iPS donor match probability. Results A total of 14 O-type haplotype homozygotes were found in 5 236 myeloid donors, which could be 49.81% (2 608/5 236), 61.29% (3 209/5 236), 79.51 Five matched levels of iPS donors were provided for the hypothetical patients with% (4 163/5 236), 75.76% (3967/536), and 97.78% (5 120/5 236); 17 HF > 1%). Conclusion The existing HLA data from China Bone Marrow Bank can create a small number of iPS cell banks with high efficiency.
