目的研究溴氰菊酯(DM)对大鼠脑组织线粒体膜通透性及细胞色素C表达的影响。方法Wistar大鼠随机分为实验组(4个不同染毒时间)和对照组,每组5只。实验组按12.5 mg/kg DM一次腹腔注射,5、24、48、72 h后处死;对照组一次腹腔注射5 mg/kg的色拉油,分别提取大鼠大脑皮层和海马的线粒体,测定线粒体膜通透性、细胞色素C氧化酶,并测定大鼠大脑皮层、海马CA1、CA2、CA3、CA4区细胞色素C的表达。结果实验组与对照组相比,5、24、48、72 h组的线粒体膜通透性增大;皮层和海马CA1区、CA2区5、24、48 h组(0.57±.04、0.67±0.09、0.58±0.04)、CA4区24 h组(0.81±0.18)细胞色素C表达增强,吸光度值的差异有统计学意义(P<0.05或P<0.01);CA2区72 h组、CA3区及CA4区5、48、72 h组细胞色素C表达,吸光度值的差异则无统计学意义(P>0.05);细胞色素C氧化酶的活力则受到抑制,差异有统计学意义(P<0.01)。结论DM能明显增加大鼠脑组织线粒体膜通透性,促使细胞色素C表达增强。
Objective To investigate the effects of deltamethrin on mitochondrial membrane permeability and cytochrome C expression in rat brain. Methods Wistar rats were randomly divided into experimental group (4 different exposure time) and control group, 5 rats in each group. The rats in the experimental group were injected intraperitoneally with 12.5 mg / kg DM for 5, 24, 48 and 72 hours, respectively. The control group was intraperitoneally injected with 5 mg / kg salad oil to separate the mitochondria of rat cerebral cortex and hippocampus respectively. Mitochondrial membrane permeability, cytochrome C oxidase, and the expression of cytochrome C in CA1, CA2, CA3 and CA4 of rat cerebral cortex and hippocampus. Results Compared with the control group, the permeability of mitochondrial membrane was increased in the 5, 24, 48 and 72 h groups. In the cortex and hippocampus, the CA2 and CA2 regions in the 5, 24 and 48 h groups (0.57 ± .04, 0.67 ± 0.09,0.58 ± 0.04). The expression of cytochrome C in CA4 area of 24 h (0.81 ± 0.18) was increased, the difference of absorbance value was statistically significant (P <0. 05 or P <0.01). There was no significant difference in cytochrome C expression and absorbance at 72 h, CA3 and CA4 in CA2 area at 5,48 and 72 h (P> 0.05) Pigment C oxidase activity was inhibited, the difference was statistically significant (P <0.01). Conclusion DM can significantly increase the permeability of mitochondrial membrane in rat brain and promote the expression of cytochrome C.
