体外培养的细胞多系非同步化细胞群体,由于群体中同时存在不同时相的细胞,这给研究射线和药物对细胞的作用机制带来一定的困难。因此,建立体外培养细胞同步化的方法对细胞学的研究具有重要的意义。本实验根据选择和诱导同步的原理,利用秋水仙胺作为中期阻断剂,对获取S_(180)-V细胞M、G_1和S各期同步化细胞的方法及其增殖规律作了初步的研究。材料与方法 s_(180)-V细胞株是余慕贞等建立的,其特点是增殖力强,半悬浮半贴壁生长,M期细胞自动浮起,G_1和S期细胞沉降贴壁。 (一)细胞培养:S_(180)-V细胞在含20%小牛血清的RPMI-1640(日本)培养液中培养
In vitro cultured multi-cell nonsynchronized cell populations, due to the simultaneous presence of cells of different phases in the population, which brings some difficulties in studying the mechanism of action of radiation and drugs on cells.
