目的:首次引入ABCG2尿酸转运蛋白,探讨其在痛风性关节炎中的作用机制和穿山龙总皂苷的干预作用。方法:通过Real-time PCR和Western blot方法,检测高尿酸血症小鼠肾脏ABCG2转运蛋白的mRNA和蛋白表达水平。结果:模型组m ABCG2的mRNA和蛋白表达量均明显降低(P<0.001)。穿山龙总皂苷低剂量组可明显回调该基因的mRNA表达(P<0.001),穿山龙总皂苷高、中剂量组均可回调ABCG2 mRNA表达(P<0.05,P<0.001)。穿山龙总皂苷高、中、低剂量组均可回调ABCG2蛋白的表达(P<0.001,P<0.05和P<0.05)。结论:穿山龙总皂苷可通过对肾脏尿酸转运体ABCG2的调控来促进尿酸的排泄实现对高尿酸血症的降尿酸作用。
OBJECTIVE: To introduce ABCG2 uric acid transporter for the first time to investigate its mechanism of action in gouty arthritis and its intervention effect. Methods: Real-time PCR and Western blot were used to detect the mRNA and protein expression of ABCG2 transporter in the kidney of hyperuricemic mice. Results: mRNA and protein expression of m ABCG2 in model group were significantly decreased (P <0.001). The mRNA expression of ABCG2 mRNA was significantly down-regulated in the low and middle dose groups (P <0.001, P <0.001). The expression of ABCG2 protein was up-regulated in high, middle and low doses of total Saponin (P <0.001, P <0.05 and P <0.05). CONCLUSION: The total saponin from S. auricula can promote uric acid excretion of hyperuricemia by regulating the kidney uric acid transporter ABCG2.
