目的 通过对胰岛素受体底物 1(IRS 1)、胰岛素样生长因子 1受体 (IGF 1R)在海马表达的观察 ,研究APP17肽对糖尿病脑病小鼠海马神经元IRS 1、IGF 1R的影响。 方法 用链脲佐菌素 (STZ)诱发小鼠糖尿病模型 ,并皮下注射APP17肽对糖尿病小鼠进行治疗 ,4周后取脑组织做IRS 1、IGF 1R免疫组织化学染色。 结果 糖尿病小鼠海马内IRS 1、IGF 1R阳性反应神经元胞浆与突起深染 ,而正常小鼠及APP17肽保护的糖尿病小鼠海马阳性细胞数目少 ,染色淡。 结论 糖尿病小鼠海马IRS 1、IGF 1R广泛表达 ;APP17肽能影响糖尿病脑病小鼠脑内IRS 1,IGF 1R的表达 ,使之接近正常
Objective To investigate the effect of APP17 peptide on IRS 1 and IGF 1R of hippocampal neurons in diabetic encephalopathy mice by observing the expression of insulin receptor substrate 1 (IRS 1) and insulin-like growth factor 1 receptor (IGF 1 R) in hippocampus. Methods Diabetic mice were induced by streptozotocin (STZ) and diabetic mice were treated with APP17 peptide subcutaneously. After 4 weeks, the brain tissues were harvested for IRS 1 and IGF 1R immunohistochemical staining. Results The numbers of hippocampal IRS-1 and IGF-1R-positive neurons in DM-treated diabetic mice were significantly higher than those in normal mice and hippocampus. The numbers of hippocampal positive cells in diabetic mice protected by APP17 peptide were less than those in normal mice. Conclusions The expression of IRS 1 and IGF 1R in the hippocampus of diabetic mice is extensively expressed. APP17 peptide can affect the expression of IRS 1 and IGF 1R in the brain of diabetic encephalopathy mice and make them close to normal
