G-CSF对不同人群CD34+细胞及T细胞亚群影响的动态变化研究

目的 探讨应用G CSF动员外周血干细胞时,异体 自体造血干细胞移植供 受者的骨髓及外周血CD3 4+ 细胞和T细胞亚群的动态变化特点及最佳外周血干细胞采集时间。方法 对12例健康供者和16例化疗7d后的恶性血液病患者,以G CSF 15 0 μg 12h ,皮下注射,连用5d。应用流式细胞仪测定骨髓及外周血CD3 4+ 细胞和T细胞亚群。结果 ①应用G CSF前,两者骨髓CD3 4+ 细胞有显著差异(P <0 0 1)。应用48h后,两者的骨髓CD3 4+ 细胞均较前有明显增加(P <0 0 1) ,两者之间仍存在明显差异(P <0 0 1)。两者的骨髓CD3 4+ 细胞高峰值出现在96h后,较应用前差异显著(P <0 0 1) ,两者之间无差异(P >0 0 5 )。②应用G CSF前,两者的外周血CD3 4+ 细胞无明显差异(P >0 0 5 )。应用72h后,两者外周血中CD3 4+ 细胞均较前有明显增加(P <0 0 1) ,两者之间无明显差异(P >0 0 5 )。96h后,两者的CD3 4+ 细胞达高峰,较应用前差异显著(P <0 0 1)。两者之间无明显差异(P >0 0 5 )。③应用G CSF对两者的T淋巴细胞亚群无明显影响。结论 应用G CSF动员外周血干细胞96h后,异体 自体造血干细胞移植供 受者的骨髓及外周血CD3 4+ 细胞均达到高峰,可适时采集外周血干细胞。 Objective To investigate the dynamic characteristics of CD3 4+ cells and T lymphocyte subsets in bone marrow and peripheral blood of allogeneic autologous hematopoietic stem cell transplantation recipients using G CSF mobilization of peripheral blood stem cells and the best time for collecting peripheral blood stem cells. Methods Twelve healthy donors and 16 patients with hematologic malignancies after 7 days of chemotherapy were injected subcutaneously with G CSF 150 μg for 5 days. Flow cytometry was used to detect CD3 4+ cells and T lymphocyte subsets in bone marrow and peripheral blood. Results ① There was a significant difference in CD3 4+ cells between the two groups before application of G CSF (P <0.01). After 48 hours of application, both CD3 4+ cells in bone marrow increased significantly (P <0.01). There was still significant difference between the two groups (P <0.01). There was no significant difference between the two groups (P> 0.05). The peak of CD3 4+ cells in both groups appeared after 96 hours, which was significantly different from that before application (P <0.01). ② Before the application of G CSF, CD3 4+ cells in peripheral blood showed no significant difference (P> 0.05). After 72 hours of application, CD3 4+ cells in peripheral blood of both groups increased significantly (P <0.01), with no significant difference (P> 0.05). After 96h, the CD3 4+ cells reached the peak, which was significantly different from that before application (P <0.01). There was no significant difference between the two (P> 0.05). ③ The application of G CSF had no significant effect on T lymphocyte subsets of both. Conclusion After mobilizing peripheral blood stem cells by G CSF for 96 hours, peripheral blood stem cells can be harvested at the peak of CD3 4+ cells in bone marrow and peripheral blood of allogeneic autologous stem cell transplantation recipients.
