目的探讨熊果酸(UA)对人肝癌SMMC-7721细胞周期的影响。方法用不同浓度的UA处理SMMC-7721细胞,采用四甲基偶氮唑盐(MTT)法测定细胞增殖的抑制作用,流式细胞仪检测细胞周期的变化,RT-PCR检测细胞周期相关基因p21、p53及增殖细胞核抗原(PCNA)的表达。结果 UA对SMMC-7721细胞生长有显著的抑制作用,且此作用呈明显的时间、剂量依赖性;UA可以诱导SMMC-7721细胞阻滞在S期,RT-PCR结果显示其能够上调p21和p53基因,下调PCNA基因。结论 UA能明显抑制SMMC-7721细胞的生长,引起细胞周期阻滞;细胞周期阻滞的发生与p21、p53、PCNA基因变化相关。
Objective To investigate the effect of ursolic acid (UA) on the cell cycle of human hepatoma SMMC-7721 cells. Methods SMMC-7721 cells were treated with different concentrations of UA, the cell proliferation was measured by MTT assay, the cell cycle was detected by flow cytometry, the expression of cell cycle related gene p21 , P53 and proliferating cell nuclear antigen (PCNA) expression. Results UA significantly inhibited the growth of SMMC-7721 cells in a dose-and time-dependent manner. UA induced SMMC-7721 cells to arrest in S phase. RT-PCR results showed that UA could up-regulate the expression of p21 and p53 Gene, down-regulate PCNA gene. Conclusion UA can significantly inhibit the growth of SMMC-7721 cells and cause cell cycle arrest. The occurrence of cell cycle arrest is related to the changes of p21, p53 and PCNA.
