目的观察异甘草素(ISL)和Flutamide联合应用对人前列腺癌细胞体外增殖的抑制作用。方法采用MTT法测定细胞增殖,流式细胞仪检测细胞凋亡,逆转录聚合酶链反应(RT-PCR)分析Bcl-2表达。结果ISL和不同浓度Flutamide联合用药比单独应用Flutamide对前列腺癌PC-3细胞增殖抑制作用明显增强(P<0.01),同时显著下调Bcl-2基因表达,增加癌细胞凋亡。结论ISL和Flutamide联合应用对前列腺癌PC-3细胞增殖抑制作用明显增强,其机制可能与下调Bcl-2基因表达及增加癌细胞凋亡有关。
Objective To investigate the inhibitory effect of isoliquiritigenin (ISL) combined with Flutamide on the proliferation of human prostate cancer cells in vitro. Methods Cell proliferation was measured by MTT assay. Apoptosis was detected by flow cytometry. The expression of Bcl-2 was analyzed by reverse transcription-polymerase chain reaction (RT-PCR). Results Compared with Flutamide alone, the inhibitory effect of ISL and Flutamide on proliferation of PC-3 cells was significantly enhanced (P <0.01). Meanwhile, the expression of Bcl-2 gene was significantly decreased and the apoptosis of cancer cells was increased. Conclusion The combination of ISL and Flutamide can significantly inhibit the proliferation of PC-3 cells. The mechanism may be related to the down-regulation of Bcl-2 gene expression and the increase of apoptosis of cancer cells.
