目的:优化五倍子总鞣质的提取工艺,为五倍子的深度开发和综合利用提供参考。方法:采用水浴加热浸提法,以没食子酸和浸出物得率为指标,在单因素试验基础上,选择提取次数、提取时间、液料比和提取温度为考察因素,采用BoxBehnken试验设计原理确定五倍子总鞣质的最佳提取工艺。结果:最佳提取工艺为加10.4倍量水于62℃水浴提取2次,每次125 min,没食子酸和浸出物得率分别为55.67%,65.11%,与理论预测值的相对偏差分别为3.30%,0.35%。结论:响应面法优选的五倍子总鞣质提取工艺方法简单、提取效率高、可预测性良好。
OBJECTIVE: To optimize the extraction process of total tannins in Galla Chinensis and to provide a reference for the further development and comprehensive utilization of Galla chinensis. Methods: Water bath heating extraction method was used to take the yield of gallic acid and extract as index. Based on the single factor test, the times of extraction, extraction time, liquid to material ratio and extraction temperature were selected as the investigation factors. The design principles of BoxBehnken were used to determine Gallic total tannins best extraction process. Results: The optimal extraction process was extraction of 10.4 times the amount of water twice in 62 ℃ water bath. The yield of gallic acid and extract was 55.67% and 65.11% respectively at 125 min. The relative deviation from the theoretical value was 3.30 %, 0.35%. CONCLUSION: The method of response surface method for total gallnut tannin extraction process is simple, high extraction efficiency, good predictability.
