目的研究回转模拟微重力(simulated microgravity,SMG)条件下成骨生长肽(OGP10-14)对MC3T3-E1成骨样细胞增殖与分化的影响。方法利用回转器模拟失重,MTT法检测OGP10-14对回转24 h,48h,72 h MC3T3-E1细胞增殖的效应,并观察培养液中碱性磷酸酶(alkaline phosphatase,ALP)、骨桥素(osteopontin,OPN)活性变化。结果回转模拟微重力条件下,10-8mol/L OGP10-14能够显著促进成骨细胞系MC3T3-E1的增殖(P<0.01)并促进成骨细胞系MC3T3-E1的ALP和OPN活性。结论回转模拟微重力条件下,OGP10-14对成骨细胞系MC3T3-E1的增殖与分化具有促进作用。
Objective To investigate the effect of osteogenic growth peptide (OGP10-14) on the proliferation and differentiation of MC3T3-E1 osteoblast-like cells under simulated microgravity (SMG). Methods The rotator was used to simulate the weightlessness. The effect of OGP10-14 on the proliferation of MC3T3-E1 cells after 24 h, 48 h, 72 h was detected by MTT assay. The alkaline phosphatase (ALP), osteopontin osteopontin, OPN) activity changes. Results Under the gyration simulated microgravity, 10-8mol / L OGP10-14 could significantly promote the proliferation of osteoblast cell line MC3T3-E1 (P <0.01) and promote the activity of ALP and OPN of osteoblast cell line MC3T3-E1. Conclusions OGP10-14 can promote the proliferation and differentiation of osteoblast cell line MC3T3-E1 under revolving simulated microgravity.
