目的探讨“双石汤”通过调控代谢综合征大鼠模型炎症因子水平,降低SOCS-3表达水平,改善MS状态的机理。方法SHR大鼠90只随机分成空白B组、中药C组、西药W组,三组再各分三小组,每小组10只;健康SD大鼠10只为正常N组。N组不做任何干预措施,SHR大鼠采取SHROB模型,时间为12周,C组造模后每日予双石汤灌胃,W组予达美康灌胃,B组不予药物干预。分别于造模后第1、2周、4周处死(B1、W1、C1)组、(B2、W2、C2)组、(N、B3、W3、C3)组,心脏取血检测TNF-α、IL-1、IL-6,取肝脏组织标本RT-PCR法检测SOCS-3 mRNA表达。结果造模后1d三组大鼠血清TNF-α、IL-1、IL-6水平均高于N组,差异有显著性意义(P<0.05),各组间差异无统计学意义(P>0.05);干预2周,三组上述因子水平无明显变化(P>0.05);干预4周,C3、W3组各因子明显下降,差异有显著性意义(P<0.05),B3组则未见明显下降(P>0.05),两两比较B3组明显高于C3、W3组,后两组间无明显差异(P>0.05)。C组大鼠的SOCS-3 mRNA表达水平低于B组,差异有显著性意义(P<0.05)。结论炎症反应在大鼠MS状态形成中起重要作用,中药双石汤可显著的降低炎症因子水平,有效防治MS状态的发生。
Objective To investigate the mechanism of Shuangshi Decoction in regulating the level of inflammatory cytokines in rats with metabolic syndrome, reducing the expression of SOCS-3 and improving the state of MS. Methods Ninety SHR rats were randomly divided into blank group B, C group and W group. Three groups were divided into three subgroups, 10 in each group. Ten normal healthy SD rats were normal N group. N group did not do any intervention, SHR rats to take SHROB model for a period of 12 weeks, C group daily modeling Shuangshi Tang gavage, W group Memecan gavage, B group without drug intervention. The levels of TNF-α and TNF-α in the blood were measured at 1, 2, 4 weeks after the model was established in the groups B1, W1, C1, B2, W2 and C2, , IL-1 and IL-6. The expression of SOCS-3 mRNA in liver tissue was detected by RT-PCR. Results The levels of TNF-α, IL-1 and IL-6 in the three groups were significantly higher than those in the N group on the 1st day after modeling (P <0.05), but there was no significant difference among the three groups (P> 0.05). After intervention for 2 weeks, there was no significant change in the above three factors (P> 0.05). After intervention for 4 weeks, the factors of C3 and W3 decreased significantly (P <0.05) (P> 0.05). There was no significant difference between the two groups (P> 0.05). The level of SOCS-3 mRNA expression in group C was lower than that in group B, the difference was significant (P <0.05). Conclusion The inflammatory reaction plays an important role in the formation of MS in rats. Chinese Shuanghuang Decoction can significantly reduce the level of inflammatory cytokines and effectively prevent the occurrence of MS.
