Geminin通过与Cdt1的相互作用抑制DNA的重复复制,保证在一个细胞周期内DNA仅被复制一次。Geminin在肿瘤细胞系及肿瘤组织中高水平表达,与肿瘤临床病理学的许多指标相关,可指示的肿瘤的预后。但当降低Geminin的表达水平时,在肿瘤细胞系出现了DNA的重复复制,细胞增殖停止,而正常细胞系的DNA复制则不受影响,细胞正常增殖。因此,以Geminin为治疗靶点能够选择性的抑制肿瘤细胞生长而不影响正常细胞,其有望成为一个理想的肿瘤治疗靶点。
Geminin inhibits DNA duplication by interacting with Cdt1, ensuring that DNA is replicated only once during a cell cycle. Geminin is expressed at high levels in tumor cell lines and tumor tissues, correlating with many of the clinical pathologies of the tumor and may indicate the prognosis of the tumor. However, when the expression level of Geminin was reduced, repeated duplication of DNA was observed in the tumor cell lines, cell proliferation was stopped, and DNA replication in normal cell lines was not affected and the cells proliferated normally. Therefore, targeting Geminin can selectively inhibit the growth of tumor cells without affecting the normal cells, which is expected to become an ideal target for tumor therapy.
