目的 研究门静脉高压症时肠系膜血管热休克蛋白 90 (HSP90 )的表达 ,并对其表达定位。方法 SD大鼠分为实验组和假手术组 ,用门静脉部分结扎法制作门静脉高压模型。 4周后取肠系膜血管组织 ,采用免疫组织化学技术、免疫印迹 (Westernblot)、逆转录 聚合酶链反应 (RT PCR)法在蛋白及mRNA水平测定门静脉高压大鼠肠系膜血管HSP90的表达。结果 门静脉高压大鼠肠系膜血管HSP90的表达增强 ,且保持在蛋白及mRNA水平上的一致。HSP90不仅在血管内膜而且还在血管平滑肌层有明显的表达。结论 HSP90可能参与了门静脉高压内脏血管病变的形成 ,从而为进一步揭示门静脉高压内脏血管病变的发病机制提供了新的思路。
Objective To study the expression and location of heat shock protein 90 (HSP90) in mesenteric vessels of patients with portal hypertension. Methods SD rats were divided into experimental group and sham operation group, and portal hypertension was made by partial ligation of portal vein. The mesenteric blood vessels were obtained 4 weeks later. The expression of mesenteric blood vessel HSP90 in portal hypertensive rats was detected by immunohistochemistry, Western blot and reverse transcription polymerase chain reaction (RT PCR) at the level of protein and mRNA. Results The expression of mesenteric vascular HSP90 in portal hypertensive rats increased and remained consistent at the level of protein and mRNA. HSP90 is expressed not only in the intima but also in the vascular smooth muscle layer. Conclusion HSP90 may be involved in the pathogenesis of portal hypertension with visceral vasculopathy, which provides a new idea for further revealing the pathogenesis of portal hypertension.
