目的:比较6种不同治法方药对肾病综合征大鼠的疗效,初步探讨其肾脏保护作用的可能机制,确立对肾病综合征大鼠的最佳治疗方药。方法:用阿霉素分次尾静脉注射法复制肾病综合征大鼠模型,实验动物随机分为8组,正常对照组、模型对照组、益气补肾组、收敛固涩组、清热利湿组、活血化瘀组、疏风通络组、综合治疗组。药物治疗组按6g生药材/200g体重给大鼠灌胃4周。测定尿蛋白、血清白蛋白、总蛋白、大鼠白细胞介素10的含量。结果:治疗4周后,益气补肾组的24h尿蛋白含量与模型组相比,显著性降低(P<0.05);血清中白蛋白含量显著性升高(P<0.05)。益气补肾组、疏风通络组、综合治疗组的IL-10含量与模型组相比,都显著性降低(P<0.05,P<0.01)。结论:在诸多治法方药中对肾病大鼠蛋白尿的疗效比较依次为益气补肾组>综合治疗组>活血化瘀组>疏风通络组>收敛固涩组,提示慢性肾小球疾病的治疗应重用益气补肾中药,益气补肾中药能够降低血清IL-10的水平,而使体液免疫的高反应状态得到调节。
OBJECTIVE: To compare the curative effect of 6 different prescriptions on nephrotic syndrome rats, to explore the possible mechanism of its protective effect on kidney and to establish the best therapeutic prescription for nephrotic syndrome rats. Methods: The model of nephrotic syndrome was duplicated with doxorubicin by tail vein injection. The experimental animals were randomly divided into 8 groups: normal control group, model control group, Yiqi Bushen group, Convergent Gusei group, Qingreli Wetness Group , Blood circulation group, Shufeng Tongluo group, comprehensive treatment group. Drug treatment group according to 6g crude drug / 200g body weight to rats for 4 weeks. Urinary protein, serum albumin, total protein, rat interleukin-10 content were determined. Results: After 4 weeks of treatment, the proteinuria of 24h of urinary albumin in Yiqi Bushen group was significantly lower than that in model group (P <0.05), and the serum albumin content was significantly increased (P <0.05). The levels of IL-10 in Yiqi Bushen Decoction, Shufeng Tongluo Decoction group and comprehensive treatment group were significantly lower than those in model group (P <0.05, P <0.01). Conclusion: The curative effect of proteinuria on nephropathy rats in many prescriptions of TCM is as follows: Yiqi Bushen group> comprehensive treatment group> Huoxuehuayu group> Shufeng Tongluo group> Convergence-Guerui group, suggesting that chronic glomerular disease Of the treatment should be re-use Qi kidney medicine, Yiqi Bushen Chinese medicine can reduce the level of serum IL-10, leaving humoral immune response to be adjusted.
