目的 :探讨卡维地洛对血管平滑肌细胞(VSMC)增殖的影响及其可能机制。方法 :通过贴壁细胞培养法建立豚鼠主动脉血管平滑肌细胞模型 ,应用 [3 H]TDR掺入试验研究卡维地洛对反应性氧族 (reactiveoxygenspecies ,ROS)、血管紧张肽Ⅱ(angiotensinⅡ ,AngⅡ )及胎牛血清 (fetalcalfserum ,FCS)诱导的血管平滑肌细胞增殖的影响。结果 :卡维地洛呈剂量依赖性地抑制ROS ,AngⅡ及FCS诱导的血管平滑肌细胞DNA合成 ,最大抑制率分别为 (48± 15 ) % ,(42± 7) %和 (44± 14) %。结论 :卡维地洛对ROS及AngⅡ诱导的血管平滑肌细胞DNA合成呈剂量相关性的抑制作用 ,提示卡维地洛可能通过抗氧化的途径抑制血管平滑肌细胞增殖
Objective: To investigate the effect of carvedilol on the proliferation of vascular smooth muscle cells (VSMCs) and its possible mechanism. Methods: The aortic vascular smooth muscle cell model of guinea pig was established by adherent cell culture. The effects of carvedilol on reactive oxygen species (ROS), angiotensin Ⅱ (AngⅡ) ) And fetal calf serum (FCS) on the proliferation of vascular smooth muscle cells. RESULTS: Carvedilol inhibited the DNA synthesis of vascular smooth muscle cells induced by ROS, AngⅡ and FCS in a dose-dependent manner with the maximum inhibition rates of (48 ± 15)%, (42 ± 7)% and (44 ± 14)%, respectively . CONCLUSION: Carvedilol can inhibit the DNA synthesis of vascular smooth muscle cells induced by ROS and Ang Ⅱ in a dose-dependent manner, suggesting that carvedilol might inhibit the proliferation of vascular smooth muscle cells through anti-oxidative pathway
