以壳聚糖为材料制备了新型的壳聚糖-促黄体激素释放激素类似物(Chitosan-LHRHA)缓释制剂。在缓释制剂埋植于实验鱼第0、2、6、14、21和30天后,采用放射免疫法、RT-PCR方法检测了实验鱼的基础生物学指标、血清睾酮(T)和雌二醇(E2)含量、芳香化酶(CYP19A)、雌激素受体(ERα)、生长激素(GH)基因mRNA的表达。结果表明:在缓释制剂埋植2~21d,壳聚糖-LHRHA缓释制剂埋植组中,实验鱼的各项生理指标显著的高于对照组和LHRHA埋植组(P<0.05)。说明壳聚糖-LHRHA缓释制剂在埋植21d内可以在实验鱼体内稳定释放,并通过正反馈途径调节鱼体血清中T和E2水平,调节鱼体生殖相关基因mRNA的表达。研究表明,壳聚糖-LHRHA缓释制剂对调节鱼类的生殖机能有良好的促进效果。
A novel chitosan-LHRHA sustained-release preparation was prepared from chitosan. After the sustained-release preparations were implanted on experimental fish at days 0, 2, 6, 14, 21 and 30, radioimmunoassay and RT-PCR were used to detect the basic biological indexes of experimental fish, serum testosterone (T) Alcohol (E2) content, aromatase (CYP19A), estrogen receptor (ERα), growth hormone (GH) gene mRNA expression. The results showed that the physiological indexes of experimental fish in the experimental group were significantly higher than those in control group and LHRHA group (P <0.05). The results showed that the chitosan-LHRHA sustained-release preparations could be released stably in the experimental fish within 21 days after implantation, and regulate the levels of T and E2 in the serum by positive feedback to regulate the mRNA expression of reproductive-related genes. Studies have shown that the chitosan-LHRHA sustained-release preparation has a good effect on regulating the reproductive function of fish.
