目的观察脐血浆(UC)对离体大鼠心肌缺血/再灌注损伤的保护作用并初步探讨其可能机制。方法取Wistar大鼠24只,随机分成正常对照组(Control)、缺血/再灌注损伤组(I/R)和脐血浆保护组(UC)。利用Langendorff离体灌流装置,复制大鼠心肌缺血/再灌注损伤模型。观察冠脉流出液中乳酸脱氢酶(LDH)活性、心肌组织匀浆中SOD活性及心肌超微结构等。结果与Control组相比,I/R组LDH活性明显增高,SOD活性降低,心肌超微结构损伤明显;脐血浆(1∶100)则明显减少LDH的漏出,同时使SOD活性升高及心肌超微结构损伤减轻。结论脐血浆能减轻大鼠心肌缺血/再灌注损伤,其机制可能与脐血浆具有抗氧化、清除自由基的作用有关。
Objective To observe the protective effect of umbilical cord blood (UC) on isolated rat myocardial ischemia / reperfusion injury and to explore its possible mechanism. Methods Twenty-four Wistar rats were randomly divided into normal control group, ischemia / reperfusion injury group (I / R) and cord blood plasma protection group (UC). Myocardial ischemia-reperfusion injury model was reproduced using Langendorff isolated perfusion device. To observe the activity of lactate dehydrogenase (LDH) in coronary effluent, SOD activity in myocardial homogenate and myocardial ultrastructure. Results Compared with Control group, LDH activity in I / R group was significantly increased, SOD activity decreased and myocardial ultrastructure damage was obvious. Umbilical plasma (1: 100) significantly decreased LDH leakage and SOD activity and myocardial ultrastructure Microstructure damage reduced. Conclusion Umbilical plasma can attenuate myocardial ischemia / reperfusion injury in rats. The mechanism may be related to the anti-oxidation and scavenging of free radicals in umbilical cord plasma.
